Rasagiline
📖 About Rasagiline
- Always check the BNF entry here for latest prescribing advice.
- MAO-B inhibitor used in the management of Parkinson’s disease.
- Approx. 5× more potent than selegiline.
⚙️ Mode of Action
- Monoamine oxidase-B (MAO-B) inhibitor.
- Reduces dopamine breakdown in the brain → prolongs dopaminergic activity.
🎯 Indication & Dose
- Indication: Parkinson’s disease, either:
– Monotherapy in early disease, OR
– Adjunct to levodopa (co-beneldopa / co-careldopa) to reduce “end-of-dose” fluctuations. - Usual dose: Rasagiline 1 mg once daily (OD).
🔀 Interactions

- ⚠️ Avoid with SSRIs, SNRIs, tricyclic antidepressants → risk of serotonin syndrome.
- ⚠️ Avoid with pethidine, tramadol, dextromethorphan (serotonergic toxicity).
⚠️ Cautions
- Use with care in hepatic impairment.
- Monitor for drug interactions (antidepressants, opioids, sympathomimetics).
🚫 Contraindications
- Severe hepatic impairment.
- Concomitant use of other MAO inhibitors (including selegiline).
💥 Side Effects
- GI: nausea, abdominal pain, dry mouth, vomiting, weight loss.
- Neuro/psych: hallucinations, depression, sleep disorders, dizziness, postural hypotension.
- Other: joint pain, dermatitis, conjunctivitis, urinary urgency, malaise, fever.
- Rare but important: impulse control disorders, melanoma risk (skin checks advised).
